Lamb stain for metachromatic granules
WebbThe granules were first described in bacteria in 1895 and initially referred too as metachromatic or volutin granules. The names derived from their property to stain red when bacteria were treated with toluidine blue, and for their presence in the bacterium Spirillum volutans , respectively. WebbMetachromatic granules are large inclusion that stores inorganic phosphate that can be used in the synthesis of ATP. In these granules, linear chains of orthophosphate are joined to each other by ester bonds. They are also known as volutin granules. They are basophilic in nature and therefore stained by basic dyes.

PRINCIPLE The Albert’s stain procedure comprises two stains; Albert’s stain 1 and Albert’s stain 2.
Metachromasia (var. metachromasy) is a characteristical change in the color of staining carried out in biological tissues, exhibited by certain dyes when they bind to particular substances present in these tissues, called chromotropes. For example, toluidine blue becomes dark blue (with a colour range from blue-red dependent on glycosaminoglycan content) when bound to cartilage. Other widely used metachromatic stains are the haematological Giemsa and May-Grunwald stains tha… Webb12 juli 2009 · The metachromatic granules, which sometimes are regarded as characteristic of the diphtherial species, are more easily differentiated when the basic …
